Maternal and perinatal forms of calcium homeostasis are in part determined by the placenta, the fetal parathyroid glands and the neonatal renal and bone systems. Alterations in these systems during the perinatal period are associated with permanent renal and skeletal abnormalities, whereas the relationship of the alterations to the maternal parathyroid glands is not clear. Recently, we have demonstrated that this maternal-fetal relationship is maintained in the perinatal period by parathyroid hormone-related protein (PTHrP). To determine the effects of perinatal PTHrP on parathyroid gland function, apoptosis and proliferation, pregnant rats were injected daily with either saline or PTHrP between embryonic day 15 and the day of delivery. Parathyroid glands from the offspring were examined for cell death and apoptosis using the TUNEL assay and Western blotting for poly (ADP-ribose) polymerase. Cell proliferation was assessed by immunohistochemical staining for Ki67, a marker for S-phase cells. The occurrence of apoptosis was increased in the parathyroid glands of the PTHrP-injected rats at the time of birth, whereas there was no difference in parathyroid cell proliferation. Both the enhanced apoptosis and cell proliferation were detected in the perinatal parathyroid glands. In the PTHrP-injected offspring, hyperphosphatemia was prevented by the treatment with the calcitriol analog, EB1089. We hypothesize that, during the perinatal period, PTHrP is a modulator of apoptotic signaling in the parathyroid gland and thereby affects the growth of the glands and the serum phosphate concentration. To help get the neighborhood on track, more than 30 people gathered at a community meeting Tuesday, Nov. 27 to develop a draft plan that covers the next six years and lists ideas for the area. “This is the time to get your input,” said Neighborhood Planner Ed Little, who joined the city’s planning team in September. “This is what we’re doing, and if you don’t like it, we’ll make it better.” The neighborhood plan, created by the city’s Local Needs Area Office, will help shape future development and serve as the basis for all future city-sponsored planning efforts for Victoria Woods. The neighborhood plan will also help provide detailed guidance to commercial entities interested in locating in the area, the city said. The plan will be updated and refined every three years, and the city is expecting it to last through at least 2024, Little said. Although the idea of neighborhood plans is relatively new in Portland, Little said it’s a common practice for major cities across the U.S. “These plans have been done all over the U.S. and other countries for quite some time,” he said. “They’re getting more and more popular.�
